heterologous expression of chit36 gene from trichoderma atroviride in canola reduces development of lesion caused by sclerotinia sclerotiorum

چکیده
an endochitinase gene (chit36) was isolated from the biocontrol fungus trichoderma atroviride. chit36 was overexpressed under the camv35s constitutive promoter in canola. transformation of cotyledonary petioles was achieved via agrobacterium tumefaciens. the insertion of transgene was verified by pcr and dna dot blotting. rt-pcr analysis indicated that the transgenic canola was able to express chit36 gene. also, we used a transgenic over-expression approach in order to investigate antifungal activity of expressed chit36 on sclerotiana sclerotiorum. antifungal activity was detected in crude protein extracts from transgenic canola using radial diffusion assay. also, lesion sizes of transgenic canola by s. sclerotiorum were significantly retarded compared to non transgenic canola plant using detached leaf assay.

The cDNA from the mycoparasitic fungus Trichoderma atroviride PTCC5220 encoding a 42 kDa chitinase (Chit42) was isolated. The nucleotide sequence of the cDNA fragment as having a 1263 bp open reading frame that encodes a 421 amino acid polypeptide, and a high homology was found withother reported Chit42 belonging to the Trichoderma sp. The 22 amino acid N-terminal sequence is a putative s...

Backgoround: Plant diseases, caused by a wide range of phytopathogenic fungi, could be managed using of Trichoderma sp, as a biocontrol agent. Cell wall degrading enzymes like chitinase from T. harzianum are important means for fungal pathogen inhibition. Overexpression of these chitinase enzymes can i...
متن کاملExpression Pattern of the Synthetic Pathogen-Inducible Promoter (SynP-FF) in the Transgenic Canola in Response to Sclerotinia sclerotiorum
Sclerotinia sclerotiorum is a phytopathogenic fungus which causes serious yield losses in canola. A pathogen inducible-promoter can facilitate the production of Sclerotinia-resistant transgenic canola plants. Inthis study, the “gain of function approach” was adopted for the construction of a pathogen-inducible promoter. The synthetic promoter technique was used, which involved the in...
متن کاملPhylogeography and Genotype-Symptom Associations in Early and Late Season Infections of Canola by Sclerotinia sclerotiorum.
ABSTRACT Both typical late season stem infections and atypical early season rosette infections of canola, a relatively new crop in the southeastern United States, were caused by Sclerotinia sclerotiorum. The 51 DNA fingerprints (from 71 isolates) did not match any fingerprints from previous studies of canola or other crops. Single locus haplotypes from nuclear DNA sequences included 18 in the i...

The epidemiology of sclerotinia rot of carrot was investigated on carrot ‘Cellobunch’ during 1999 and 2000 in Ontario. Apothecia were first detected in the crop in early August to mid-September, after the carrot canopy closed and after 7–11 days with soil matric potentials between –0.1 and –0.4 bars and soil temperatures between 14 and 23 °C. Ascospores were first detected in mid-July to mid-Au...
